Just as a point of information, that's the Tuolumne Hikers' Shuttle - operated by Delaware North for a fee. They have assorted one-way and round trip (on one bus) prices from Yosemite Valley. Although it's designed for hikers, it can be taken as a tour bus, with the driver pointing out points of interest. They can theoretically pick someone up, but it's rather hard to meet the schedule. Last year I took it from the Valley to Tenaya Lake. It was supposed to be $14 (I didn't do the maximum one-way trip to Tuolumne Meadows) but ended up paying $16 at the tour desk.

There's also a free shuttle service that's scheduled to meet every stop each half hour from 7 AM to 7 PM along Tioga Road. It goes from Tuolumne Meadows to Oldstead Point and back.
